Description

Description: This assay employs a two-site sandwich ELISA to quantitate ZMYND19 in samples. An antibody specific for ZMYND19 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ZMYND19 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZMYND19 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZMYND19 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The deduced 227-amino acid protein contains a C-terminal zinc finger motif that shares a high degree of amino acid identity to the zinc finger domains of myeloid translocation protein-8 (CBFA2T1), Nervy, DEAF1 (MYND) proteins. ZMYND19 contains multiple putative phosphorylation sites. The deduced human, mouse, and rat ZMYND19 proteins share 100% amino acid identity. Immunocytochemistry of transfected HEK293 cells localized ZMYND19 to the cytoplasm and MCHR1 to the plasma membrane; however, ZMYND19 colocalized to the plasma membrane when cotransfected with MCHR1. Northern blot analysis of human tissues detected a 1.5-kb transcript in all areas of the brain, especially cerebellum, and in heart, liver, skeletal muscle, kidney, testis, placenta, and stomach
